- ItemAntibiotic resistance of Escherichia coli isolated from Ukrainian Betula verrucosa Ehrh. pollen after microbiological analysis(Slovak Agricultural University in Nitra, 2013-08-01) Shevtsova, Tetiana; Hleba, Lukáš; Kačániová, Miroslava; Brindza, Ján; Garkava, Kateryna; Гаркава, Катерина ГригорівнаSeven samples of silver birch pollen from different habitat of Ukraine were investigated in order to estimate their contamination with the Enterobacteriaceae family, anaerobic bacteria and fungi. Also resistance of 108 strains of Escherichia coli isolated from seven samples of Ukrainian Betula verrucosa Ehrh. pollen against 5 antibiotics: ampicillin, chloramphenicol, meropenem, ceftriaxone and ofloxacin were determined. Disc diffusion method was used for antibiotic suceptibility testing according to EUCAST 2012. It is established the concentrations of enterobacteria ranged from 0.00 to 4.16 log cfu/g, of anaerobic bacteria – 2.48 to 4.90 log cfu/g and concentration of fungi ranged from 2.48 to 4.14 log cfu/g. Degree of pollen contamination is different depending on the habitats. The resistance of E. coli isolates was determined against ampicillin, chloramphenicol, meropenem and ofloxacin. But intermediate resistance in the 33.3% of E. coli isolates and susceptibility in the 8.3% to ceftriaxone was found out. Antibiotic resistance was evaluated for all samples of pollen in whole.

- ItemApplication of immunoglobulin-binding proteins A, G, L in the affinity chromatography(2014) Васильченко, Ольга Анатоліївна; Sviatenko, O.V.; Gorbatiuk, O.B.Proteins A, G and L are native or recombinant proteins of microbial origin that bind to mammalian immunoglobulins. Preferably recombinant variants of proteins A, G, L are used in biotechnology for affinity sorbents production. Сomparative characteristics of proteins A, G, L and affinity sorbents on the basis of them, advantages and disadvantages of these proteins application as ligands in the affinity chromatography are done. Analysis of proteins A, G, L properties is presented. Binding specificities and affinities of these proteins differ between species and antibody subclass. Protein А has high affinity to human IgG1, IgG2, IgG4, mouse IgG2a, IgG2b, IgG3, goat and sheep IgG2, dog, cat, guinea pig, rabbit IgG. Protein G binds strongly to human, mouse, cow, goat, sheep and rabbit IgG. Protein L has ability of strong binding to immunoglobulin kappa-chains of human, mouse, rat and pig. Expediency of application of affinity chromatography with usage of sorbents on the basis of immobilized proteins A, G, L are shown for isolation and purification of antibodies different classes. Previously mentioned method is used as an alternative to conventional methods of protein purification, such as ion-exchange, hydrophobic interactions, metal affinity chromatography, ethanol precipitation due to simplicity in usage, possibility of one-step purification process, obtaining of proteins high level purity, multiuse at maintenance of proper storage and usage conditions. Affinity sorbents on the basis of immobilized proteins A, G, L are used not only for antibodies purification, but also for extraction of different antibodies fractions from blood serum.

- ItemBiotechnological aspects of hydrocarbons biodegradation(2012) Васильченко, Ольга Анатоліївна; Aliieva, O.R.; Matvyeyeva, O.L.; Salata, A.M.In review the present situation of petroleum pollution, its consequences and basic oil composition are described. The up to date survey of hydrocarbon degradation microbiology and major strains are presented with respect to following practical introduction of obtained data. A general characteristic of oil hydrocarbons biodegradation is done. Biodegradation metabolism of petroleum products different categories and its biochemistry are described. Principal biochemical pathways are indicated. Biotechnological properties of hydrocarbon degrading microorganisms including that of industrial significance are investigated. Much attention is given to biological and environmental factors that influence completeness and rate of hydrocarbons biodegradation. It was established that microorganisms consortium biodegradation is faster and more complete that single species ones. The effects of hydrocarbons presence in medium on biochemical and cytological levels of microbial metabolism are also analyzed. The major methods of microbial resistance to petroleum products toxic influence are described. It is shown that one of the most important among them is metabolic processes shifting towards surfactants.
- ItemBIOTECHNOLOGICAL ASPECTS OF POLY-β-HYDROXYBUTYRATE BIOSYNTHESIS(2011) Vasylchenko, O. A.; Yastremskaya, L. S.; Salata, A. M.; Salata, O. S.; Fedun, N. O.In the review polyhydroxyalkanoates characteristic, including poly β hydroxybutyrate (P3HB) which is a polymer produced by microorganisms (Ralstonia eutropha — superproducer) is done. Biochemical pathways and conditions of P3HB microbial synthesis have been examined. Thermoplastic features of microbial P3HB in comparison with classic chemical polymers (polyethylene and polypropy lene) have been analyzed. It was shown that according to its properties it was the most similar to syn thetic polypropylene in comparison to other biopolymers, and it could be its good substitute. Microbial P3HB ability for biodestruction to CO2 and H2O has shown as extremely perspective pro perty for this polymer usage in different industry branches and for environment saving against anthro pogenous pollution.
- ItemChanges in expression of BetV1 allergen of silver birch pollen in urbanized area of Ukraine(Taylor & Francis Group, 2013) Žiarovská, Jana; Labajová, Mária; Ražná, Katarína; Bežo, Milan; Śtefunová, Veronika; Shevtsova, Tetiana; Garkava, Kateryna; Brindza, JánThe aim of the study was to determinate the level of expression of silver birch allergen Betv1 in pollen samples from different Ukraine areas by RT-qPCR SYBR Green assay. Protocol for quantifying the expression of Betv1 allergen was developed when testing of three housekeeping genes—cyclophylin, alpha-tubulin and transcription factor CBF1. Samples from urbanized area was analysed by real-time PCR when a sample from forest growth conditions was used as a calibrator. Real-time PCR based quantifying of Bet v 1 provides a useful method for rapid and sensitive analyses of this silver birch allergen. Our results show higher expression levels in samples from central parts of urbanized area as housing estates when compared to the samples from borders of the urbanized area.
- ItemChem. listy 108, 10801083(2014) Laboratorní přístroje a postupy 1080 antioxidačná aktivita peľu briez vo vzťahu k expresii betv1 alergénu(2014) Гаркава, Катерина Григорівна; Žiarovská, Jana; Shevtsova, Tetiana; Gažo, Ján; Brindza, Ján; Bežo, MilanThe aim of the study was to determine the total anti-oxidant activity and expression of silver birch allergen BetV1 in pollen samples from various Ukraine areas. The obtained results show higher expression of BetV1 allergen in samples from central parts of urbanized areas (ca. 1.5 times) compared with the samples from borders of the urbanized areas (ca. 0.55 times). The total antioxidant ac-tivity of aqueous pollen extracts ranged from 80.5 % to 85.5 %, for ethanolic extracts 60.3–84.9 %. Positive corre-lation was found between antioxidant activity in ethanolic pollen extract and BetV1 allergen expresion.

Oil products in sewer water are in the dissolved and emulsified condition. For the removal of such contaminations fraction it is expedient to use the process of biosorption before a bioscrubbing. The exception of oil products from effluents is expedient to carry out in pinotenk, that is a device with gas-liquid reflow. Processes of biosorption of contaminations and their partial oxidization by the organisms of active silt are taking place in pinotenk

The study is focused on the comparative analysis among pollen samples when complex of biotic, abiotic and anthropogenic factors in different places of growth affect. The obtained results are compared to the existing requirements. It is established excess of limits on the content of cadmium, lead, chromium, the concentrations of enterobacteria and mesophilic aerobic bacteria in some samples of birch pollen. The Tukey test was used to determine the differences between the means at a level of P < 0.05. Also in this work, the steps from collection to storage of wind-pollinated pollen were analyzed.

- ItemCryopreservation: a way to preserve the genetic resources of animals(National aviation university, 2021-06) Цимбалюк, Анастасія Олексіївна; Tsimbaliuk, Anastasiiа OleksiivnaIntense manmade pressure on ecosystems leads to a reduction in biological diversity. At present, deepfreezing of biomaterials cryopreservation is widely used to preserve species diversity and duplicate valuable specimens from world collections. Intense anthropogenic pressure on ecosystems leads to a reduction in the natural capacity for self-extraction of species and limits their reproduction. The number and diversity of valuable and rare, but lowresistant species, is declining; they are replaced by a few inferior or harmful but highly productive and resistant species (gray rat, gray crow, etc.). A significant number of fish, birds, animals (Araripsky Manakin, Red Monkey, Spotted Harlequin, Red–finned Blueeyed, Java Rhinoceros) were on the verge of extinction. Each species has a unique genotype, and a rapid decline in numbers leads to a reduction in genetic diversity, making it vulnerable to overfishing, environmental pollution and accompanied by depression with consequent extinction. The steady decline in biodiversity threatens the fact that the degradation processes hidden in natural populations appear suddenly and lead to the complete extinction of the most common species. Such dynamics are growing like a landslide, and the number of such economically and ecologically valuable fish species as the nelma, muksun population in the Siberian sturgeon has been catastrophically reduced. Their ecological niches are occupied by more stable and less in demand carpets, rattan and others. For sustainable development, modern and future selection, the most important tasks are maintenance of species diversity, conservation of valuable species, development of approaches and organization of longterm storage of the gene pool. Slow freezing, a common method for cryopreservation of embrions, causes osmotic shock (solution effect) and intracellular ice crystallization leading to cell damage. Vitrification is an alternative method for cryopreservation in which cells are exposed to a higher concentration of cryoprotectants and frozen with an ultrarapid freezing velocity, resulting in an ice crystal free, solid glass-like structure. Presently, vitrification is a popular method for cryopreservation of embryos. However, vitrification of oocytes is still challenging due to their complex structure and sensitivity to chilling.

To evaluate the prospects of therapeutic effect of the created drug on the rate of post-stress recovery of native mechanokinetic characteristics of the muscular system.
- ItemEFFECT OF ALFALFA MEDICAGO SATIVA ON VITAL ACTIVITY OF YEAST S. CEREVISIAE(2018) Решетняк, Людмила Расулівна; Семенова, О.І.; Коломоєць, Н.А.Purpose: improvement of biotechnological qualities of baker’s yeast promotes the intensification of fermentation processes, semis maturation, and ultimately the improvement of the quality of final product. Yeasts do not always satisfy the requirements. Therefore studies that are aimed at the intensification of fermentative activity and improvement of the quality of yeasts are interest. The problem of examination of stimulating effect of substances from different nature and origin on the growth and quality of yeasts is not new. One of the methods of intensification of the technological process of yeast production is selection of culture medium by replacement of mineral source of nitrogen with plant-based source. In our opinion, it is suitable to use for this purpose. Methods: in this study, we examined the ability of yeast cells to utilize organic nitrogen from a plant-based source of protein such as alfalfa granules. The use of this method allowed us to carve out the optimal components of the nutrient medium, which has provided the yeast with improved quality indicators. Results: we examined the effect of alfalfa as a source of nitrogen on the vital activity of yeast S. cerevisiae. Discussion: Alfalfa granules can be used in a cultural medium for barm yeast cultivation on a production scale.
- ItemEffect of prebiotics on lyzosymic activity of the Bacillus genus bacteria(2013) Васильченко, Ольга Анатоліївна; Saveka, М.A.Beneficial effects on a host health of probiotic strains of spore-forming bacteria of the Bacillus genus in combination with prebiotic substance lactulose were investigated. The influence of prebiotic lactulose on the Bacillus subtilis 39 and Bacillus subtilis 51 lyzosymic activity were analyzed. The dependence of the growth inhibition zones extension on lactulose quantity introduced into cultural medium was displayed.
