- ItemAntibiotic resistance of Escherichia coli isolated from Ukrainian Betula verrucosa Ehrh. pollen after microbiological analysis(Slovak Agricultural University in Nitra, 2013-08-01) Shevtsova, Tetiana; Hleba, Lukáš; Kačániová, Miroslava; Brindza, Ján; Garkava, Kateryna; Гаркава, Катерина ГригорівнаSeven samples of silver birch pollen from different habitat of Ukraine were investigated in order to estimate their contamination with the Enterobacteriaceae family, anaerobic bacteria and fungi. Also resistance of 108 strains of Escherichia coli isolated from seven samples of Ukrainian Betula verrucosa Ehrh. pollen against 5 antibiotics: ampicillin, chloramphenicol, meropenem, ceftriaxone and ofloxacin were determined. Disc diffusion method was used for antibiotic suceptibility testing according to EUCAST 2012. It is established the concentrations of enterobacteria ranged from 0.00 to 4.16 log cfu/g, of anaerobic bacteria – 2.48 to 4.90 log cfu/g and concentration of fungi ranged from 2.48 to 4.14 log cfu/g. Degree of pollen contamination is different depending on the habitats. The resistance of E. coli isolates was determined against ampicillin, chloramphenicol, meropenem and ofloxacin. But intermediate resistance in the 33.3% of E. coli isolates and susceptibility in the 8.3% to ceftriaxone was found out. Antibiotic resistance was evaluated for all samples of pollen in whole.
- ItemChanges in expression of BetV1 allergen of silver birch pollen in urbanized area of Ukraine(Taylor & Francis Group, 2013) Žiarovská, Jana; Labajová, Mária; Ražná, Katarína; Bežo, Milan; Śtefunová, Veronika; Shevtsova, Tetiana; Garkava, Kateryna; Brindza, JánThe aim of the study was to determinate the level of expression of silver birch allergen Betv1 in pollen samples from different Ukraine areas by RT-qPCR SYBR Green assay. Protocol for quantifying the expression of Betv1 allergen was developed when testing of three housekeeping genes—cyclophylin, alpha-tubulin and transcription factor CBF1. Samples from urbanized area was analysed by real-time PCR when a sample from forest growth conditions was used as a calibrator. Real-time PCR based quantifying of Bet v 1 provides a useful method for rapid and sensitive analyses of this silver birch allergen. Our results show higher expression levels in samples from central parts of urbanized area as housing estates when compared to the samples from borders of the urbanized area.

The obtained results show higher expression of BetV1 allergen in samples from central parts of urbanized areas (ca. 1.5 times) compared with the samples from borders of the urbanized areas (ca. 0.55 times). The total antioxidant ac-tivity of aqueous pollen extracts ranged from 80.5 % to 85.5 %, for ethanolic extracts 60.3–84.9 %. Positive corre-lation was found between antioxidant activity in ethanolic pollen extract and BetV1 allergen expresion.
- ItemContamination of Betula verrucosa Ehrh. pollen by microorganisms, mycotoxins and heavy metals(Faculty of Biotechnology and Food Sciences of SAU, 2014-06-01) Shevtsova, Tetiana; Kačániová, Miroslava; Garkava, Kateryna; Brindza, Ján; Petrova, Jana; Гаркава, Катерина ГригорівнаPollen samples of silver birch from Ukraine and Slovakia were investigated in order to estimate their contamination with heavy metals (Cd, Pb, Hg, Cr, As, Ni, Se, Co), representatives of the Enterobacteriaceae family, mesophilic aerobic bacteria, fungi and mycotoxins. The study is focused on the comparative analysis among pollen samples when complex of biotic, abiotic and anthropogenic factors in different places of growth affect. The obtained results are compared to the existing requirements. It is established excess of limits on the content of cadmium, lead, chromium, the concentrations of enterobacteria and mesophilic aerobic bacteria in some samples of birch pollen. The Tukey test was used to determine the differences between the means at a level of P < 0.05. Also in this work, the steps from collection to storage of wind-pollinated pollen were analyzed.
- ItemValidation of HMG CoA Reductase as Internal Control for Hazelnut Pollen Allergens Expression Analysis(2015-02-13) Garkava, Katerina; Žiarovská, Jana; Nikolaieva, Natalia; Brindza, Ján; Гаркава, Катерина ГригорівнаIn the study, 3-hydroxy-3-methylglutaryl coenzyme A reductase gene was validated for its application in hazelnut pollen allergens CorA and profillin expression analysis. HMG CoA reductase was used as internal control in expression analysis by one-step real-time PCR approach. Pollen sample from rural area from Sumy region (Ukraine) was used as the calibrator and the sample from Kamenets-Podolskiy (cement plant area; Ukraine) was chosen for the expession analysis. Using one-step real time PCR approach, following values of expression level were obtained in this study: 0,63 x higher for CorA and 51,6 higher x for profilin hazelnut pollen ellergen. The results and parameters of expression profile data are comparable to the two-step real-time PCR, that were previously reported for Corylus avelana, L. pollen allergens and confirmed HMG CoA reductase as suitable internal control for the CorA and profillin expression analysis.
